Dear Dr. Cheng,
thank you for a great talk. I had a question about the use of ICC in the microbiome studies.
on slide 42, it looks like the ICC was used to determine the reproducibility of the results by performing duplicate extractions from several individuals (to assess the extraction technique). this application of ICC makes sense to me.
in contrast, on slide 45, it sounds like a longitudinal study was performed testing diversity over time within the same individuals. In this setting, I am not sure I understand the use of ICC unless it is known that diversity is very stable in the same person of many months.
on slide 51, it sounded like there was detection of stool microbe diversity changes over the summer months, so my question is that I do not understand why the ICC is applied to the results of a given person's stool samples over time if a normal finding is to see a change in diversity over time? What is the ICC telling you in this instance?
thank you
