Access Question - cleaning up string text

I realize this is an access question in a biostat forum, but I've exhausted alternatives and hoping someone on here can help. 

Here's  my issue:

I had 2 columns of incomplete data in an access document, I was able to combine the data into 1 column, unfortunately the column contains a lot of string text. I want to clean up the string text so the column only reads "positive". I've tried the update query command, with the replace function without success. 

Any other suggestions on how to do this?
